Exploration of a Versatile G-Quadruplex Dual-Point-Of-Care Detection System for Biosensing
point-of-care detection (POCT) for vital biomolecules is of great significance for home testing in biomedical and clinical diagnosis due to the unique detection advantages with rapid, convenient, sensitive, non-damaging, and specific identification. Based on the advantages of the excellent designability, hydrophilicity, biocompatibility of the G4 motif, to date, a considerable achievement was obtained by colorimetric signal output (single-POCT) based on G4 sensing. However, there are still exist inherent defects of single-POCT , such as insensitivity to tiny changes in analyte concentration and substantial interference from the intrinsic color of the measured sample or ambient light, leading to false positives. Here, we introduced G4-Hemin into the temperature panel to achieve the first G4 dual-POCT (colorimetric and temperature discoloration sticker) system . The versatility has been demonstrated in detecting Ochratoxin A, ATP, and thrombin by adjusting different modules, and the practicality has been verified for actual samples detection by the naked eye and temperature discoloration sticker. It could not only well eliminate the disadvantages of single signal output, but also significantly broaden the practicality and scope of applications for G4 sensing. Advances in convenience and accuracy of color change with the reaction solution and temperature discoloration sticker in dual-POCT, this G4 biosensor system holds significant potential in POCT for home testing and biological emergency response
